离子对凝胶色谱分离头孢菌素中高分子杂质实验条件的选择 被引量:13

摘要 以建立Ceftazidime中高分子杂质含量的测定方法为例,对不同离子对试剂、洗脱剂pH对头孢菌素在离子对凝胶色谱中色谱行为的影响,洗脱剂类型、离子对浓度等因素对头孢菌素中高分子杂质的分离效果的影响进行了探讨。 所建立的测定Ceftazidime中高分子杂质的离子对凝胶色谱法为:内径1.3cm、Vt=50ml的Sephadex G-10(粒度40~120μ)凝胶柱:洗脱剂采用pH7.0的0.4mol/L磷酸缓冲液或含0.25mol/L(NH_4)_2SO^4,pH7.0的0.01mol/L磷酸盐缓冲液,洗脱速度为0.5ml/min。上样量为200μl。 For establishment of a method to be used in the assay of high moieo-ulat weight impurities from ceftazidime,the effects of different iou-pair. reagents and eluants with different pH value on the chromatographic behavour of cephalosporins, the effects of eluants with different ioa-pair reagents and with different concentration of the reagents on the performence of gel filtration chromatographic sy.stern were discussed. The method established was as follows. Column, diameter was 1.3 cm, Vt = 50ml. packing Sephadez G-10(40 - 120uuuuuuuuu) eluant. pH 7.0, 0.4mol/L Phosphate buffer or 0.25 mol/L ammonium sulfate,pH 7.0, 0.01 mol/L Phosphate buffers flow rate. 0.5 ml/ min; sample volume; 200u1.
