整合素β1对牙龈卟啉单胞菌诱导牙龈上皮细胞金属基质蛋白酶-2,9表达的影响 被引量:2

MMP-2 and MMP-9 expression induced by porphyromonas gingivalis in gingival epithelial cell was influenced by integrin β1 expression

摘要 目的本研究探讨整合素β1对牙龈卟啉单胞菌诱导牙龈上皮细胞MMP-2和MMP-9表达的影响。方法采用Western blot和q RT-PCR检测牙龈卟啉单胞菌感染前后牙龈上皮细胞MMP-2和MMP-9的表达;通过integrinβ1-si RNA下调牙龈上皮细胞整合素β1的表达,采用Western blot和q RT-PCR检测牙龈卟啉单胞菌感染对照组和整合素β1干扰组的牙龈上皮细胞后MMP-2和MMP-9表达。结果牙龈卟啉单胞菌感染牙龈上皮细胞可以诱导牙龈上皮细胞MMP-2和MMP-9的表达增加,并且表达都是在12 h时达到最高峰。下调牙龈上皮细胞整合素β1后,牙龈卟啉单胞菌诱导si RNA-integrinβ1干扰组牙龈上皮细胞MMP-2和MMP-9表达明显减少,与对照组相比存在明显差异(P<0.01)。结论整合素β1在牙龈卟啉单胞菌诱导牙龈上皮细胞MMP-2和MMP-9表达中起很重要的作用,可能成为治疗牙周炎的新靶点。 Objective This study explores integrinβ1 in the role of MMP-2 and MMP-2 expression induced by porphyromonas gingivalis in gingival epithelial cell. Methods MMP-2 and MMP-9 expression was measured by Western blot and qRT-PCR in gingival epithelial cell before and after porphyromonas gingivalis infection. Next, integrinβ1 was downregulated in gingival epithelial cell by integrinβ1-siRNA, Western blot and qRT-PCR was used to determine the MMP-2 and MMP-9 expression in control and integrinβ1-siRNA group infected with porphyromonas gingivalis. Results MMP-2 and MMP-9 expression was increased after porphyromonas gingivalis infection, both at peak in 12 h. MMP-2 and MMP-9 expression in integrinβ1-siRNA group after porphyromonas gingivalis infection was decreased significantly compared with control (P〈0.01, respectively). Conclusion MMP-2 and MMP-9 expression in gingival epithelial cell after porphyromonas gingivalis infection can be mediated by integirnβ1 and this may be new target to treat periodontitis.
