摘要
赤霉素属于植物激素,主要成份为A_3。我们建立了用反相HPLC分析测定A_3含量的方法,及时反馈发酵液、吸附废液、解吸液及固体粗品中赤霉素效价情况,为中间体各环节的质量控制提供了简便、快速、重现性良好的方法。使用Waters液相仪,层析柱C_(18)(150×4.6mm),流动相为无水甲醇、水、磷酸混合液,标准曲线相关系数0.9996,回收率100.5%,CV<1%。
Gibberellins was plant hormones. Among these components in the complex, the major activity component was As.
To determine the concentration of A3 in Gibberellins, a reverse phase HPLC was established for the analysis of Gibberellins. The feed back in potency of Gibberellins among cultrue liquid and adsorbent eluate was ra-pidly performed. A simple, fast, accurate, sensitive method was provided for quality control of various link of intermediate.
By means of Waters Liquid Chromatography system, ODS C18 column (150#4.6 mm). The mobile phase was a mixture of 34% methanol, 66% water, added a little phosphate with UV detector at 215nm.
Standard curves was linear in the range between 90 and 800 μg GA3/ml.The correlation coefficient was better than 0.999. The average recovery was 100.5%, CV< 1%.
The result was quite satisfactory.
