摘要
用反相高效液相色谱柱前衍生法测定葡萄酒样品中的氨基酸组成,对方法参数进行了优化。结果表明,流速对分离效果有明显影响,流动相pH在中性偏碱范围内对分离效果无明显影响。荧光和紫外检测联用可以有效的对24种氨基酸进行定性和定量检测,23种氨基酸检出限在3~6 pmol。葡萄酒中氨基酸总含量为9 666.3μmoL/L,其中人体必需氨基酸含量为5 138.2μmoL/μL。18种氨基酸的加标回收率在85.5%~114.4%之间。
The amino acids in wine was determined by precolumn derivatization of RPHPLC to optimize method parameter.Results show that,flow velocity has obvious influence on separation effect,and there is no obvious influence of mobile phase pH on separation effect with the scope of neutral to alkaline.FLD and DAD are used for qualitative and quantitative detection of 24 amino acids,the detection limit of 23 amino acids is 3~6 pmol.The total content of amino acids in wine is 9 666.3μmol/L,thereinto the content of essential amino acid for human body is5 138.2μg moL/μL.The recovery of 18 amino acids is 85.5%~114.4%.
