SiRNA抑制CDK6表达阻滞大肠癌细胞增殖和细胞周期进展

摘要 目的:探讨Si RNA抑制CDK6表达后对大肠癌细胞生长及周期的影响。方法:实时荧光定量PCR检测CDK6在大肠癌与大肠组织中的表达差异。Si RNA转染抑制CDK6表达,MTT和流式细胞仪被用来检测细胞增殖和细胞周期改变。Western blot检测细胞周期因子表达的改变。结果:与大肠组织相比,CDK6基因在大肠癌中表达明显增高。下调CDK6表达后,大肠细胞生长能力明显降低,且细胞周期也出现G1/S转换障碍。机制分析显示,在抑制CDK6表达后,细胞增殖相关因子CDK4和CCND1下降,而P21表达明显提升。结论:CDK6作为候选癌基因通过调节CDK4、CCND1及P21表达影响大肠癌细胞增殖以及细胞周期转化。
