摘要
为优化甘薯[Ipomoea batatas(L.)Lam.]茎叶中绿原酸超声提取工艺,在单因素试验的基础上,选择乙醇溶液体积分数、提取时间、料液比(g:m L,下同)3个对绿原酸提取率影响较大的因素,采用BoxBehnken中心组合试验设计对各因素进行优化组合,运用Design Expert 8.05b软件进行响应面回归分析,建立了绿原酸提取率的二次多项式回归方程。得到的优化组合条件为乙醇溶液体积分数70%、料液比1∶20、超声时间24 min,绿原酸提取率为3.097 2%,与响应面拟合所得方程的预测值3.106 8%相近。试验所用甘薯茎叶价廉易得,提取方法高效可行,分析方法可信,可为甘薯茎叶的开发利用提供依据。
In order to optimize the ultrasonic extraction technology of chlorogenic acid from leaves and stems of Ipomoea batatas (L.) Lam.,on the base of single-factor experiment,it selected the following three factors which greatly influnence the extraction ratio of chlorogenic acid: the volume fraction of ethanol solution,the extraction time and solid-liquid ratio. Then, it adopted the combination experiment design of Box-Behnken centre to optimize those factors, used Design Expert 8.05b soft- ware to analyze the response surface regression and established quadratic polynomial regression equation of the extraction ratio of chlorogenic acid. The optimum combination condition obtained were the follow spects: the volume fraction of ethanol solu- tion was 70%, solid-liquid ratio was l:20(g:mL),extraction time was 24 min and the extraction yield of chlorogenic acid was 3.097 2%, which was well consistent with the predicted value of the response surface 3.106 8%. The extraction method has the advantage of time-saving, high efficiency and good reliablity of analysis method,which can provide the basis for the de- velopment and utilization of the leaves and stems of Ipomoea batatas(L.) Lam..
