摘要
目的探讨外用他克莫司联合窄谱中波紫外线(NB-UVB)对白癜风豚鼠的白细胞介素10(IL-10)水平及酪氨酸酶水平的影响。方法建立白癜风动物模型,按体重随机分成6组,每组8只,正常组、模型组、对照组(白斑区,外用凡士林,2次/天)、单用药物组(白斑区,外用0.03%他克莫司,2次/天);单用光照组(白斑区,单用NB-UVB照射,隔日1次)、联合组(白斑区,外用他克莫司联合NB-UVB照射)均连续2个月。给药结束,用ELISA法检测给药及照射前后豚鼠皮肤组织匀浆及血清中IL-10含量的变化,用比色法检测白癜风豚鼠给药及照射前后血清中酪氨酸酶水平的变化。结果与给药及照射前比较,联合组的血清及组织匀浆中IL-10含量明显增高(P<0.01),但血清中酪氨酸酶水平无明显变化。结论联合应用NB-UVB与他克莫司可明显提高白癜风豚鼠血清及组织匀浆中的IL-10水平。
Objective To investigate the effect of combined narrow band ultraviolet B(NB-UVB) and tacrolimus on the changes in the level of tyrosinase and interleukin-10(IL-10) in Guinea pig with vitiligo.Methods Animal models with vitiligo were built first.And then all the animal models were randomly divided into six groups of eight according to the principle of weight balance:normal group,model group,control group(2 times per a day),medication alone group(0.03% tacrolimus ointment skin smear,2 times per a day),light alone group(white spot area use NB-UVB irradiation,once every other a day),combination group(topical tacrolimus combination NB-UVB irradiation,frequency,method and time same to medication alone group and light alone group).Colorimetric method and ELISA method were used to detect the changes in the level of tyrosinase in serum and IL-10 in skin tissue homogenates and serum respectively,before and after the administration and irradiate.Results No significant changes in the level of tyrosinase in serum were observed before and after combined administration of NB-UVB and tacrolimus in animal models with vitiligo.While IL-10 contents in skin tissue homogenates and serum were significantly higher than that before administration,and the difference was statistically significant(P 〈 0.01).Conclusion Combined treatment of NB-UVB and tacrolimuscan significantly improve the level of IL-10 in skin tissue homogenates and serum for animal models with vitiligo.
