摘要
甾体激素主要包括肾上腺糖皮质激素、雌激素、孕激素、雄激素和维生素D,临床常用的甾体激素类药物多为其衍生物或人工合成品。细胞色素P450(CYP450)代谢酶系在甾体激素的生物合成及代谢清除过程中起着关键的作用,而甾体激素也可通过对CYP450的诱导或抑制而影响其酶蛋白表达及代谢活性,二者之间的相互作用决定了临床多种疾病(如肿瘤、炎症或自身免疫性疾病)发生的易感性及药物疗效。本文对甾体激素类药物与细胞色素P450代谢酶系相互作用的研究进展进行阐述。
Steroid hormones are mainly consisted of glucocorticoid,estrogen,progesterone,androgen and vitamin D.The common clinical used steroid hormone drugs are mainly the derivatives or synthetic compounds of the steroid hormones.Cytochrome P450(CYP450),a super family of the heme-thiolate proteins,plays a vital role in both the synthetic and metabolic process of the steroid hormones.However,the steroids could also influence the mRNA expression and metabolic activity of the CYP450.Consequently,the interaction between the steroids and CYP450 may determine the human ’s susceptibility to many diseases such as tumour,inflammation or autoimmune diseases and can also influence the drug’s therapic effect.In this review,we introduce the recent progress in the interaction between the steroid hormones and CYP450.
