枳实中活性单体环氧橙皮油素抗溃疡性结肠炎的作用

Effect of Epoxyaurapten from Aurantii Fructus Immaturus on Treating Ulcerative Colitis

目的:观察环氧橙皮油素对溃疡性结肠炎小鼠治疗作用。方法:BALB/C小鼠分为6组,分别为环氧橙皮油素低、中、高剂量组(80,160,320μmol·L-1),正常组,柳氮磺砒啶肠溶片组(520 mg·kg-1)和葡聚糖硫酸钠(DSS)模型组。除模型组外均按0.01 m L·g-1体重剂量ig给药,正常组给予等体积的生理盐水。除正常组外各给药组在给药同时自由饮用4%DSS水溶液,连续造模10 d。观察各组药物对BALB/C小鼠疾病活动指数的影响;采用结肠损伤评分法,观察各组药物对病变肠组织的影响;ELISA法测结肠组织细胞因子肿瘤坏死因子-α(TNF-α),白细胞介素-10(IL-10)含量;Western blot法测定环氧橙皮油素对受损结肠平滑肌肌球蛋白轻链激酶(MLCK)表达水平影响。结果:与模型组比较,环氧橙皮油素中、高剂量组疾病活动指数明显降低(P<0.05),结肠损伤指数明显降低(P<0.05),环氧橙皮油素各剂量组结肠组织TNF-α含量逐渐降低,其中高剂量组与模型组有显著差异(P<0.05),环氧橙皮油素各剂量组结肠组织IL-10含量逐渐升高,其中中、高剂量组与模型组均有显著差异(P<0.05),环氧橙皮油素各剂量组均能使受损结肠平滑肌细胞肌球蛋白轻链激酶表达量降低,中、高剂量组与模型组有显著性差异(P<0.05)。结论:环氧橙皮油素通过不同作用机制对溃疡性结肠炎起到治疗作用。

Objective： To observe the effects of epoxyauraptene from Aurantii Fructus Immaturus on ulcerative colitis （UC） in BALB/C mice. Method： BALB/C mice were divided into 6 groups： the normal group （normal saline）, the model group （normal saline）, the sulfasalazine group （520 mg ·kg -1）, the low-, medium- and high-dose epoxyaurapten groups （80, 160, 320 μmol ·L-1）. The UC model was induced in mice by receiving 4% dextran sulphate sodium salt solution for 10 days. All mice received 0.01 mL·g -1 of the corresponding medicines. Disease activity index of BALB/C mice was observed. Colonic mueosa damage repair in mice was evaluated by Colon injury scoring method. The contents of tumor necrosis factor α （TNF-α） and interleukin-lO （IL-10） in BALB/C mice colon tissue were determined by using ELISA. Colon smooth muscle myosin light chain kinase expression level was determined by Western blot. Result： Compared with the model group, the disease activity index decreased in the medium-and high-dose epoxyaurapten groups （P 〈 0.05 ） , colonic injury index decreased, the content of TNF-α in mice tissue decreased in the high-dose epoxyaurapten group （P 〈 0.05 ） , the level of IL-10 increased, the damaged colon smooth muscle myosin light chain kinase expression decreased in the medium-and high-dose epoxyaurapten groups （P 〈 0. 05 ）. Conclusion： Epoxyaurapten has a good effect in treating UC by various kinds of mechanisms in mice.
