摘要
目的 采用液质联用技术和紫外检测法,对蒙药栀子的化学成分进行结构鉴定,为其药理作用的进一步研究及临床应用提供科学依据。方法 色谱分离采用Venusil MP C18色谱柱(4.6 mm×300 mm,5μm);流动相为0.1%甲酸水溶液和甲醇,梯度洗脱;体积流量为1.0 m L/min;质谱定性采用多级离子阱质谱,负离子模式扫描;紫外检测范围为210~400 nm。结果 在优化后的液质联用条件下,通过液相色谱/离子阱质谱(LC/MSD Trap)法检测出栀子中的13个成分。经文献查阅,化合物3、5、6、8、11为首次经液质联用从栀子中得到。结论 LC/MSD Trap联用技术为分离和鉴定栀子中的化学成分确定一种快速、科学的分析方法。
AIM To analyze the structures of chemical constituents from traditional mongolian medicine Gardenia jasminoides Ellis by LC-MSD Trap and UV,providing scientific basis for further study and clinical application of its pharmacological activities. METHODS Venusil MP C18Column( 4. 6 mm × 300 mm,5 μm) was used. The mobile phase consisted of 0. 1% formic acid in water and methanol was used in gradient elution; the flow rate was1. 0 m L / min; multistage electrospray ion trap mass spectrometry was applied for qualitative analysis under negative ion mode; the UV detection range was 210- 400 nm. RESULTS Under optimized conditions,13 constituents in Gardenia jasminoides Ellis were identified by liquid chromatography / ion trap mass spectrometry( LC / MSD Trap).Through the literature search,compounds 3,5,6,8,11 were firstly isolated from Gardenia jasminoides Ellis by this method. CONCLUSION A fast and scientific analysis method is established,which can isolate and identify the chemical constituents of Gardenia jasminoides Ellis.
