Cry1Ab蛋白在果蝇体内的富集作用及对其体内保护酶活性的影响

摘要 为了研究Cry1Ab蛋白对非靶标生物的生态安全性,通过采用酶联免疫技术(ELISA)测定了果蝇体内的Cry1Ab蛋白含量,运用紫外分光光度法检测了超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、乙酰胆碱酯酶(ACh E)的活性。结果表明:在前10天内Cry1Ab蛋白在果蝇体内不断累积,果蝇体内的ACh E、SOD和GSH-Px酶活性与对照相比均受到抑制,SOD和GSH-Px酶活性的抑制程度随其体内毒蛋白含量的增高而降低,ACh E酶活性的抑制程度则随之而升高;但10天后果蝇体内Cry1Ab蛋白含量逐渐降低,其体内3种酶的活性均随体内毒蛋白含量的降低而逐渐升高,但仍低于对照。该研究表明Cry1Ab蛋白在果蝇体内具有富集效应,并且其体内Cry1Ab蛋白含量对其体内SOD、GSH-Px、ACh E具有抑制作用。 To study the effects of Cry1 Ab protein on ecology security of non-target organisms, enzyme-linkedimmunosorbent assay(ELISA) was applied to test Cry1 Ab protein of Drosophila melanogaster and ultravioletspectroscopy was applied to test the activities of superoxide dismutase(SOD), glutathione peroxidase(GSH-Px)and acetylcholinesterase(ACh E) in Drosophila melanogaster. The results showed that Cry1 Ab proteincontinuously accumulated in the first 10 days. Compared with the normal level, specific activity of ACh E, SODand GSH-Px were inhabited. The inhibition on the activity of ACh E was strengthened, and the activities ofGSH- Px and SOD were reduced with the increasing content of Cry1 Ab protein. After 10 days, content ofpoisonous protein was reduced, inhibitions on ACh E, SOD and GSH-Px were reduced, but activities of thethree enzymes were still lower than those of the control groups. These results suggest that there is bio-concentration of Cry1 Ab protein and specific activity of ACh E, SOD, GSH-Px can be inhabited.
