摘要
目的通过观察白花前胡甲素(praeruptorinA,PdIa)对低氧引起的大鼠肺动脉平滑肌细胞基础钙及钙池操纵性钙内流(storeoperatedcalciumentry,SOCE)上调的影响,以探讨Pd-Ia是否参与低氧时细胞内钙稳态的调节。方法原代培养肺动脉平滑肌细胞。将细胞分为常氧组、缺氧组及缺氧+Pd-Ia组,采用双波长测定法测定细胞基础钙及SOCE。结果缺氧可使细胞基础钙浓度及SOCE升高或增加[(220.61±14.79)nmol/LVS(103.35±11.68)nmol/L;(270.02±22.66)nmol/Lvs(56.41±9.04)nmol/L,P值均〈0.01]。与缺氧组相比,缺氧+Pd-Ia组细胞的基础钙浓度及SOCE降低或减少[(132.19±8.35)nmol/L;(112.56±21.97)nmol/L,P值均〈0.01]。结论Pd—Ia可以明显抑制低氧引起的大鼠肺动脉细胞基础钙及SOCE的上调。
Objective In this study,a primary culture system for the rat distal pulmonary arterial smooth muscle cell (PASMC) was established to observe the effect of praeruptorin A (Pd- I a) treatment on the basal intracellular calcium concentration ([Ca2+]i) and store operated calcium entry (SOCE) in rat PASMC. Methods We employed the primary culture method for the rat distal PASMC including the enzymatically dissociation of PASMC from the freshly isolated distal pulmonary artery,and the culture of PASMC were divided into Normoxia group, Hypoxia group and hypoxia + Pd- I a group. The InCyte system was used to measure the basal [Ca2+]i and SOCE after substantial treatment. Results Compared with Normoxia group, the basal [Ca2+ ]i and SOCE both significantly increased in hypoxia group [(220. 61±14.79) nmol/L vs (103.35 ±11. 68) nmol/L; (270.02 ±22.66) nmo[/L vs (56.41 ±9.04) nmol/L,both P 〈0.01]. Compared with Hypoxia group, the the basal [Ca2+ ]i and SOCE were both significantly reduced in Hypoxia+ Pd- Ia grou[( 112.56 ± 21.97) nmol/L; (132.19± 8.35 ) nmol/L, both P〈0.01]. Conclusions Pd- I a inhibit chronically hypoxic enhancement ofbasal [Ca2+ ]i and SOCE in rat distal pulmonary arterial smooth muscle cells.
