Measure of Total Polyphenols Content in Zhaotong Apples by Folin-Ciocalteu Method

[目的]采用Folin-Ciocalteu法测定苹果总酚含量,探索基层实验室快速准确测定苹果总酚的有效方法,为昭通苹果生产及加工工艺提供科学依据。[方法]采用70%乙醇酸性溶液作溶剂,35℃下超声提取苹果总酚,以Folin-Ciocalteu试剂和120 g/L Na2CO3溶液处理,在765 nm处测其吸光度。[结果]以没食子酸为标准,质量浓度在0-125.4 mg/L范围内与吸光度线性关系良好（r=0.999 1）,据此测定昭通苹果试样的总酚含量范围为361.1-1 008.0 mg/kg,加标回收率试验结果为103.2%,RSD 1.4%。[结论]该方法操作简便,重复性好,结果准确可靠,适用于基层实验室。

[ Objective ] In order to provide seientific basis for production and processing of apples in Zhaotong, as well as finding convenient and exact method of measuring the total polyphenol in apples, the total polyphenol eontent was measured by Folin-Ciocalteu assay. [ Method ] Using 70% acid ethyl alcohol as the solvent, total polyphcnols of apples was extracted by supersonic method at 35 ℃, then added Folin-Ciocalteu reagent and 120 g/L Na2CO3 solution, and measured the absorbancy at 765 nm. [ Result] Basing on gallic acid, the mass concentration had an obvious linear relation with absorbency within the scope of 0 - 125.4 mg/L （ r = 0. 999 1 ）. As a consequence, the total polyphenols content in samples is 361.1 - 1 008.0 mg/kg, and adding standard recovery result is 103.2% , RSD 1.4%. [ Conclusion] This method is convenient, repeatable and faithful, suitable for basic level laboratory.
