TLR-9单核苷酸多态性与鲍氏不动杆菌感染相关性研究 被引量:1

Association of single-nucleotide polymorphism in TLR9 receptors with Acinetobacter baumannii infection

摘要 目的探讨鲍氏不动杆菌感染和TOLL样受体成员TLR9单核苷酸多态性之间相关性。方法根据编码序列同义或非同义突变、频率>5%的SNPs位点以及文献报道与细菌感染相关SNPs位点作为候选SNPs位点,采用iMLDR技术对401例鲍氏不动杆菌感染患者(试验组)和385例非感染患者(对照组)的血标本进行TLR-9受体SNP位点检测,对各基因位点基因型频数、等位基因频数及单倍型分布进行比较和分析。结果在试验组和对照组均衡性检验中,鲍氏不动杆菌感染与性别差异无统计学意义,但是t检验显示与年龄增加相关,因而遗传分析以年龄做校正;TLR-9基因位点rs187084,不符合哈迪-温伯格平衡;此位点在经年龄校正后的加性模型分析和隐形模型分析中,显示此位点多态性与鲍氏不动杆菌感染呈相关性。结论 TLR-9基因SNPs位点rs187084可能与中国人群鲍氏不动杆菌感染存在相关性,但由于不符合哈迪-温伯格平衡,还需要增加随机样本量进一步证实。 OBJECTIVE To determine the associations between the TLR‐9 single‐nucleotide polymorphism (SNP) and the Acinetobacter baumannii infection .METHODS The candidate SNPs at TLR‐9 were selected according to their frequency (more than 5% ) and position ,as well as their associations with infectious diseases reported previ‐ously .The TLR‐9 SNPs were detected in the blood specimens that were obtained from 401 patients with A .bau‐mannii infection and 385 patients without infections by using improved multiplex ligation detection reaction (iM‐LDR ) technique .The genotype frequency and allele frequency at the gene loci as well as the distribution of haplo‐type were observed and compared .RESULTS The equilibrium test for the case group and the control group showed the A .baumannii infection was not associated with the genders ,however ,the t‐test demonstrated that it was asso‐ciated with the increase of age ,thus ,the age was used to adjust the genetic analysis .The SNPs (rs187084) of TLR‐9 gene was not in line with Hardy‐Weinberg equilibrium .The additive model analysis and invisible model a‐nalysis of the locus ,adjusted by the age ,indicated that the polymorphism was associated with the A .baumannii infection .CONCLUSION The SNPs (rs187084) of TLR‐9 gene may be associated with the A .baumannii infection in Chinese population ,and it is necessary to increase the random sample size to further prove due to its inconformi‐ty with Hardy‐Weinberg equilibrium .
