变异型分化抗原簇44(17)对宫颈癌HeLa细胞、SiHa细胞系增殖的影响 被引量:2

摘要 目的探讨变异型分化抗原簇44(17)(CD44v17)对人宫颈癌HeLa细胞、SiHa细胞系增殖的影响。方法用lipofectamine 2000将CD44v17siRNA、pc DNA3.1-CD44v17质粒及其阴性对照组转染HeLa细胞、SiHa细胞,通过CCK-8、划痕实验、流式细胞术、免疫印迹法(Western blotting)、细胞周期测定方法等观察CD44v17对人宫颈癌细胞系HeLa细胞及SiHa细胞的增殖、凋亡、迁移等能力的影响。结果 CCK-8法证实CD44v17促进宫颈癌细胞的生长;划痕实验显示CD44v17可增强宫颈癌细胞的迁移能力;流式细胞术及Western blotting检测凋亡相关蛋白验证了CD44v17具有抑制宫颈癌细胞凋亡的作用;细胞周期测定显示干扰CD44v17 RNA后,S期细胞比例降低,G1期细胞增高,细胞阻滞于G1期并诱导细胞凋亡。结论 CD44v17能抑制宫颈癌细胞凋亡,促进宫颈癌细胞的生长,增强宫颈癌细胞的增殖迁移能力,它有可能成为宫颈癌治疗的新靶点。 Objective:To explore the effect of CD44v17 on proliferation of human cervical cancer Si Ha cells and HeLa cells.Methods:Lipofectamine 2000 was used to transfect CD44v17 siRNA,pc DNA3.1-CD44v17 plasmid and negative controls into Si Ha cells and HeLa cells; CCK-8 method,wound healing test,flow cytometry,Western blotting,cell cycle analysis were used to observe the effect of CD44v17 on proliferation,apoptosis,and migration of human cervical cancer Si Ha cells and HeLa cells.Results:CCK-8 method confirmed that CD44v17 could promote proliferation of human cervical cancer Si Ha cells and HeLa cells; wound healing test showed that CD44v17 could enhance migration of human cervical cancer Si Ha cells and HeLa cells; flow cytometry and Western blotting verified the inhibiting effect of CD44v17 on apoptosis of human cervical cancer Si Ha cells and HeLa cells; cell cycle analysis showed that after interfering CD44v17 RNA,the proportion of cells at S phase decreased,while the proportion of cells at G1 phase increased,the cells were arrested at G1 phase and cell apoptosis was induced.Conclusion:CD44v17 can inhibit apoptosis,promote proliferation,enhance cell migration of human cervical cancer Si Ha cells and HeLa cells; CD44v17 may be a new target for treatment of cervical cancer.
