矽肺壹期患者血清中核因子E2相关因子2和8-异前列腺素F2α水平及意义 被引量:3

Expression levels and significances of serum Nrf2 and 8-iso-PGF2α in patients with stageⅠsilicosis

摘要 目的探讨矽肺壹期患者血清中核因子E2相关因子2(Nrf2)和8-异前列腺素F2α(8-iso-PGF2α)水平变化及意义。方法采用酶联免疫吸附法检测67例矽肺壹期患者(矽肺组),38名井下健康接尘矿工(接尘对照组)及30名无粉尘接触的行职业健康检查的人员(非接尘对照组)血清中Nrf2和8-iso-PGF2α水平;用肺功能机检测用力肺活量、一秒钟用力呼气量、1秒率和最大通气量的实测值占预计值百分比共4项肺功能指标。结果矽肺组人群Nrf2和8-iso-PGF2α水平均低于接尘对照组(P<0.05),接尘对照组人群Nrf2水平高于非接尘对照组(P<0.05);肺功能异常亚组矽肺壹期患者血清8-iso-PGF2α水平低于肺功能正常亚组(P<0.01);矽肺壹期患者血清Nrf2水平与8-iso-PGF2α水平呈正相关(Speraman相关系数为0.869,P<0.01),但Nrf2、8-iso-PGF2α水平与上述4个肺功能指标均无相关性。结论 Nrf2和8-iso-PGF2α水平变化与矽肺壹期发生有关。 Objective To explore the expression level changes and the significances of serum nuclear factor-E2-related factor 2( Nrf2) and 8-iso-prostaglandin F2α( 8-iso-PGF2α) in the patients with stageⅠ silicosis. Methods The levels of Nrf2 and 8-iso-PGF2α were determined by enzyme-linked immunosorbent assays in serum of 67 stage Ⅰ silicosis patients( silicosis group),38 healthy underground miners( dust-exposed control group),and 30 healthy controls( unexposed control group); the proportion of actual measured value of forced vital capacity( FVC),forced expiratory volume in one second( FEV1),FEV1/ FVC,and maximal ventilatory volume were determined by special pulmonary function testing machine. Results The level of Nrf2 and 8-iso-PGF2α was significantly lower in silicosis group than that in dust-exposed control group( P〈0. 05),the level of Nrf2 in dust-exposed control group was significantly increased compared with that of unexposed control group( P〈0. 05); the level of 8-iso-PGF2α in stage Ⅰ silicosis patients with abnormal pulmonary function was significantly lower than that of silicosis patients with normal lung function( P〈0. 01). In stage Ⅰ silicosis patients,there was positive correlation between 8-iso-PGF2α and Nrf2( Spearman correlation coefficient was 0. 869,P〈0. 01),but no correlation was found between Nrf2,8-iso-PGF2α and lung function indexes. Conclusion The expression level changes of Nrf2 and 8-iso-PGF2α are related with the occurrence of stageⅠ silicosis.
