瓦尼木层孔菌发酵液提取物对人结肠癌LoVo细胞增殖的影响 被引量:2

摘要 桑黄(Phellinus spp.)是一种珍贵的多年生大型药用真菌,具有独特的抗癌功效,是目前国际公认的生物治癌药剂中效率最高的一种药用真菌。该文从区域特种食(药)用真菌的开发利用角度出发,以采自黑龙江完达山的杨树桑黄即瓦尼木层孔菌(Phellinus vaninii)为研究对象,采用MTT方法比较了水提和酯提方法菌丝发酵液4个提取物对人结肠癌Lo Vo细胞增殖的抑制效果,计算高效提取物最佳作用浓度和IC50,绘制加药时间与抑制Lo Vo细胞增殖效果的曲线。研究结果表明,不同处理方法获得的提取物对人结肠癌Lo Vo细胞增殖的抑制效果差异显著,其中酯提效果好于水提,在1 000μg·m L-1浓度条件下,加药48 h后,正丁醇提取样品对人结肠癌Lo Vo细胞抑制率最高为86.8%,乙酸乙酯提取样品抑制率为87.5%,超声波提取样品对人结肠癌Lo Vo细胞抑制率为58.3%,直接水提效果最差,只有57.8%;桑黄菌丝体发酵液乙酸乙酯提取物IC50为53.459μg·m L-1,在此浓度下,加药后96 h抑制率达到90%以上,加药120 h抑制率达95%,抑制率也随着加药时间的增加而提高。这为探求瓦尼木层孔菌资源价值的科学评价与有效开发奠定了基础,具有重要的学术和现实意义。 Phellinus spp. is a valuable, perennial and macro medicinal mushroom with a unique anticancer efficacy and is one of the most efficient and internationally recognized medicinal mushroom among the biological drugs in the cure of cancer. From the view of the development and utilization of regional special edible-medicinal mushroom, taking the PheUinus vaninii collected from the region of Wanda Mountains in Heilongjiang province as the research object, the inhibition effect of 4 mycelium fermented broth extracts on the proliferation of colon cancer LoVo cells by water extract and ethyl acetate extract with the MTT assay was compared. The best concentration and IC50 of efficient extract were calculated and the administration time and inhibi- tion effect of proliferation of LoVo ceils were determined. The results showed that the difference of the inhibiting effect of the extracts obtained by different methods on the proliferation of colon cancer LoVo ceils was significant. Among them, the effect of ethyl acetate extract was better than water extract. At the concentration of 1 000 μg·mL-1 and 48 h after the medicine, the high- est inhibition rate of butanol extract samples on colon cancer LoVo cells was 86.8%, which of ethyl acetate extract samples was 87.5%, the inhibition rate of ultrasonic bath extract was 58.3% and the effect of direct water extract was the worst, only 57.8%. The IC50 of the ethyl acetate extract of mycelium fermented broth of Phellinus vaninii was 53.459 μg·mL-1. At the concentration and 96 h after the medicine, the inhibition rate was more than 90%, 120 h after the medicine, the inhibition rate was 95% and increases with the medicine time. It laid the foundation for seeking the scientific evaluation and the effective development of resources value of PheUinus vaninii and had an important academic value and realistic importance.
