血必净注射液对系统性红斑狼疮小鼠脾脏T淋巴细胞的免疫调节作用 被引量:1

Effect of Xuebijing injection(血必净注射液)on immune function of T lymphocytes in systemic lupus erythematosus in mice

摘要 目的:探讨血必净注射液对系统性红斑狼疮(SLE)小鼠T淋巴细胞的免疫调节作用。方法:以清洁级EB病毒膜抗原BLLF1基因转基因小鼠30只为SLE模型小鼠,随机分为模型组(8周龄和10周龄各5只)、8周龄治疗组(10只)和10周龄治疗组(10只)。两治疗组给予血必净注射液6.4ml/kg腹腔内注射,每日1次,连续14d。治疗14d后处死,分离脾脏,提取DC和T淋巴细胞,荧光定量PCR和酶联免疫吸附试验检测T细胞孵育上清和脾组织匀浆液IL-2和TNF-α基因及细胞因子表达水平。结果:与正常对照组比较,SLE小鼠IL-4、TNF-α含量显著升高,而IL-2、IFN-γ水平明显降低,脾脏组织及T淋巴细胞IL-2的基因表达显著下降,而TNF-α基因表达则明显升高(P<0.05或P<0.01)。给予血必净注射液治疗后,与模型组比较,狼疮鼠IL-4、TNF-α含量显著降低,而IL-2、IFN-γ水平明显升高,IL-2基因表达显著升高,TNF-α基因表达明显降低,差异具有统计学意义(P<0.05或P<0.01)。两个治疗组之间比较差异无统计学意义(P>0.05)。结论:血必净治疗能够将脾脏的Th2细胞转化为Th1细胞,改变Th1/Th2的比例和机体的免疫应答类型,对系统性红斑狼疮的免疫功能产生影响。 Objective:To observe the effect of Xuebijing injection(血必净注射液)on T lymphocyte and its potential mechanisms in systemic lupus erythematosus(SLE).Methods:A widely used mouse model,SLE-prone BLLF1 mice aged 8-10 weeks,was employed.Xuebijing injection in a dose of 6.4ml/kg was given via the intraperitoneal administration in the Xuebijing treatment group.Spleen sample was divided as following:one part was used to procure DC with MACS microbeads and T cell by using column of nylon wool,and the other part was used to determine gene and protein expressions of various cytokines.Cells were cultured and the contents of cytokines released into supernatants were determined.The mRNA expression was measured by realtime quantitative PCR.All cytokines were determined in supernatant by ELISA kits for mice.Results:In the model control group,the levels of IL-4and TNF-αwere increased significantly,but the levels of IL-2and IFN-γwere decreased markedly.The gene expressions of IL-2were also decreased and TNF-αwere increased.Xuebijing treatment could significantly down-regulate the release of IL-4,TNF-αand decrease in IL-2,IFN-γ.Conclusions:Treatment with Xuebijing injection might influence the polarization of T cells in animals with SLE and induced Th cells to drift to Th1 cells.
