二十碳五烯酸的发酵工艺

Production of Eicosapentaenoic Acid by Fermentation of Yarrowia lipolytic

摘要 以产二十碳五烯酸(EPA)的解脂耶罗维亚酵母(Yarrowia lipolytic)SIPI-N-5为出发菌株,通过碳源和氮源优化,以5%甘油为碳源,0.5%酵母浸粉为氮源,摇瓶中EPA的产量为1 324 mg/L。进一步在5 L发酵罐进行分批发酵培养,采用较适宜的培养基并补加葡萄糖和酵母浸粉,EPA产量提高至2 701 mg/L。提取发酵产物,获得占总油脂含量33.9%的EPA。 A Yarrowia lipolytic SIPI-N-5 strain, which could produce eicosapentaenoic acid (EPA), was used as the starting strain. After optimization of carbon and nitrogen sources, the titer of EPA was 1 324 mg/L in fermentation medium consisting of 5 % glycerol and 0.5 % yeast extract in flask shake culture. Furthermore, fed-batch culture of SIPI-N-5 was conducted in a 5 L fermentor. The yield of EPA was increased to 2 701 mg/L in an appropriate medium by feeding glucose and yeast extract during the fermentation process. The fermentation product was extracted and EPA equivalent to 33.9 % of total lipids was obtained.
