Relationship between the Chemical Compositions of Tween-80 and Its Hemolysis

摘要 采用体外溶血试验考察,对组成Tween-80的8类化学成分进行致溶血性安全评价。结果显示,它们的致溶血能力有显著差异。聚氧乙烯山梨醇酐(PS)、聚氧乙烯异山梨醇(PI)和聚氧乙烯异山梨醇二油酸酯(PID)在浓度为500μg/ml时未出现溶血,聚氧乙烯山梨醇酐三油酸酯(PSTri)浓度为300μg/ml、聚氧乙烯山梨醇酐四油酸酯(PSTetra)浓度为500μg/ml时可导致溶血,而聚氧乙烯山梨醇酐一油酸酯(PSM)浓度为80μg/ml、聚氧乙烯山梨醇酐二油酸酯(PSD)浓度为120μg/ml、聚氧乙烯异山梨醇一油酸酯(PIM)浓度为30μg/ml时即可导致溶血。提示应控制市售Tween-80中各化学组分的含量,以减少其溶血毒性。 In vitro hemolysis test was conducted to determine the hemolysis caused by eight types of chemical compositions of Tween-80. The results showed that there were significant differences among the hemolysis of the eight compositions. No hemolysis was observed in polyoxyethylene sorbitan (PS), polyoxyethylene isosorbide (PI) or polyoxyethylene isosorbide dioleates (PID) at 500μg/ml. Polyoxyethylene sorbitan trioleates (PSTri) at 300 μg/ml and polyoxyethylene sorbitan tetraoleates (PSTetra) at 500μg/ml could cause hemolysis, while polyoxyethylene sorbitan monooleate (PSM) at 80 μg/ml, polyoxyethylene sorbitan dioleates (PSD) at 120 μg/ml and polyoxyethylene isosorbide monooleate (PIM) at 30 μg/ml showed hemolytic effect. It was suggested that the contents of chemical compositions of commercially available Tween-80 should be controlled in order to reduce the hemolytic toxicity.
