摘要
成纤维细胞生长因子-21(FGF-21)是FGF家族的一员。现有研究表明,FGF-21是一种血糖调节因子,有望被开发成为治疗糖尿病的候选药物。本实验是在前期研究的基础上,对高效表达FGF-21基因的工程菌株进行接种、发酵、诱导表达,将表达产物纯化,PEG修饰提高其体内半衰期,通过改变反应体系的反应时间、反应温度、蛋白浓度和反应物之间的质量比优化修饰反应。阴离子交换层析纯化反应混合物,最终得到纯度大于98%的活性FGF-21蛋白。
Fibroblast growth factor 21( FGF-21) is a member of the FGF family. The existing studies show that,FGF-21 is a blood sugar regulator and is expected to be developed as a drug for diabetes. The study is to express FGF-21 in sumo prokaryotic expression system based on the previous study. The expression product is purified,PEG-modified to improve its in vivo half-life. The reaction is optimized by varying the time,the temperature,protein concentration and the ratio of the reactants. The mixture was purified by ion exchange chromatography,and the purity of FGF-21 is over 98%.
