香猪肺源性枯草芽孢杆菌的分离鉴定及体外抑菌作用

Isolation,Identification and Bacteriostasis of Pulmonary Bacillus subtilis in Guizhou Congjiang Xiang Pig

为获得猪呼吸系统的益生菌,以对疾病具有较强抗性的从江香猪为对象,从肺组织中分离到1株细菌Bp-1并对其进行形态学观察和生理生化特性鉴定,经克隆测序分析16SrDNA核苷酸序列和二级结构的相似性,利用琼脂扩散法和混菌拮抗培养法研究Bp-1对大肠杆菌、金黄色葡萄球菌和猪链球菌3种致病菌的抑制作用。结果表明:菌株Bp-1与枯草芽孢杆菌(Bacillus subtilis)位于同一分枝,相似性达100%;与已知枯草芽孢杆菌菌株168的16SrRNA可变区二级结构相比无明显差异,因此将Bp-1鉴定为枯草芽孢杆菌。枯草芽孢杆菌Bp-1的液体培养物对3种致病菌均有抑制作用;对大肠杆菌的拮抗作用较强,对金黄色葡萄球菌的拮抗作用较弱。枯草芽孢杆菌Bp-1可作为猪呼吸系统的益生菌对其深入研究。

The morphology and physiological-biochemical characteristic of the bacterium strain Bp-1isolated from lung samples of Congjiang Xiang Pig with a strong disease resistance was identified,then the similarity of 16 SrDNA nucleotide sequence and secondary structure was analyzed by clone sequencing,and finally the inhibition effect of strain Bp-1on Escherichia Coil,Staphylococcus aereus and Streptococcus suis was determined by agar diffusion and mixed bacteria antagonism culture methods to obtain the probiotics of swine respiratory system.Strain Bp-1and B.subtilis are in the same sub-branch and the similarity is up to 100%.Strain Bp-1is identified as B.subtilis because there is no obvious difference in the secondary structure of 16 SrRNA variable region between strain Bp-1and B.subtilis strain 168.The liquid culture substance of strain Bp-1 has the inhibition effect on three pathogenic bacteria,the strong antagonism against E.Coil and the weak antagonism against S.aereus.Strain Bp-1should be used as a probiotic of swine respiratory system for further study.
