摘要
为探明新疆地区绵羊肺炎支原体(Mycoplasma ovipneumoniae,MO)流行株是否存在遗传多样性,用KM2培养基对塔城地区7个羊场疑似MO感染的肺脏病料进行病原分离,并进行菌落形态、生化试验和分子生物学鉴定。结果表明:15株分离均为MO,菌落呈乳头状,菌体呈多形性;能水解葡萄糖但不能水解精氨酸、尿素,可使美兰牛乳褪色、吸附并引起红细胞溶血;16Sr RNA序列大小为361bp。其中,5株MO HSP70基因的核苷酸同源性为94.1%,氨基酸同源性为93.2%,与MO标准毒株Y98相比,分离株在593位点均插入Gln(谷氨酰胺)。MO新疆分离株已发生遗传变异,存在遗传多样性。
The colonial morphology,biochemical indexes and molecular biology of the pathogens isolated from lung samples of 7suspected M.ovipneumoniaeinfection sheep farms were analyzed to discuss genetic diversity of M.ovipneumoniaeepepidemic strain in Xinjiang.Results:5isolated strains all are M.ovipneumoniae.The colony is papillary and thallus is polymorphism.15 isolated strains can hydrolyze glucose,but can not hydrolyze arginine and urea.15 isolated strains can fade and adsorb methylene blue milk,which results in erythrocytes hemolysis.The size of 16 Sr RNA sequence is 361 bp.The nucleotide homology and amino acid sequence homology of HSP70 gene among 5isolated strains are 94.1% and93.2%respectively.No.593 locus of the isolated strains is inserted with Gln compared with the standard strain Y98.There is genetic diversity in the isolated strains in Xinjiang.
