摘要
为建立苦楝皮药材中川楝素UPLC的测定方法,并评价不同来源苦楝药材的质量,为苦楝皮药材的临床使用提供参考,采用超高效液相色谱法对不同来源苦楝皮中的川楝素进行测定。结果表明:在色谱柱为Agilent ZORBAX RRHD Eclipse Plus C18(2.1×100mm,1.8μm),流动相为乙腈-水(34∶66),流速为0.10mL/min,检测波长为210nm,柱温为30℃时,川楝素在0.044 8~0.784 0μg的线性良好,平均加样回收率为97.03%,RSD小于3%(n=6)。不同来源苦楝药材中川楝素的含量差异较大,同部位(茎皮)中川楝素的含量差异达10倍以上,同植株不同部位的药材川楝素的含量以根皮〉茎皮〉枝皮的顺序排列。
To establish the determination methods and evaluate quality of different sources of toosendanin in M.toosendan and M.azedarach,so as to provide a reference for clinical use,UPLC-DAD method was employed in the determination.Results:The separation was performed on Agilent ZORBAX RRHD Eclipse Plus C18column(2.1×100mm,1.8μm)with the mobile phase acetonitrile-water(34:66),flow rate 0.10mL/min,detection wavelength 210 nm and the column temperature 30℃,toosendaninhad a good linear in 0.044 8~0.784 0μg,the average recovery was 97.03%,RSD was less than 3%(n=6).The contents of toosendaninfrom different sources were quite different,differences in toosendanin were up more than 10 times in content sites(bark),contents in different parts followed the sequence of root bark〉bark〉 branch bark.
