摘要
目的研究不同强度恒定磁场(CMF)对小鼠成骨细胞增殖及功能的影响。方法选取小鼠成骨细胞MC3T3-E1细胞株进行培养,加载0、50、100、150、200 m T的恒定磁场,筛选出最显著促进、抑制成骨细胞生长的加载强度(150、200 m T)。将培养的细胞随机分为3组,分别加载0 m T(对照组)、150 m T(促进组)、200 m T(抑制组)的CMF,检测细胞增殖活性、Ⅰ型前胶原羧基端肽(PICP)、碱性磷酸酶(ALP)、骨钙素(OCN)含量。结果 150 m T的CMF可以促进小鼠成骨细胞增殖及功能,强度为200 m T的CMF可以抑制小鼠成骨细胞的增殖及功能。结论 CMF可以促进小鼠成骨细胞增殖及功能,且存在"强度窗"效应,超过这一特定强度时,则表现为抑制作用,150 m T左右即为可能的"强度窗"之一。
Objective To investigate the effects of different-intensity constant magnetic fields (CMFs) on the proliferation and function of osteoblasts in mice. Methods The mice osteoblast cell line MC3T3-E1 were selected to culture, then stimulated by the CMFs with 0, 50, 100, 150 or 200 mT, and screened the intensity which promote and inhibit the growth of osteoblasts the most significantly (150, 200 mT). MC3T3-E1 were divided into three group, and then stimulated by the CMFs with 0, 150, 200 mT, respectively. The proliferation rate of osteoblasts, PICP expression, ALP activity, OCN activity were evaluated. Result The CMFs with 150 mT can promote the mice osteoblast proliferation and function, while the CMFs with 200 mT can inhibit the mice osteoblast proliferation and function. Conclusion The CMFs can promote the mice osteoblast proliferation and function, and has ‘intensity window’ effect. The CMFs beyond the certain level showed inhibitory effect, and thus 150 mT may be one of the possible ‘intensity window’.
