摘要
目的:探讨穿山龙总皂苷对反复脑缺血再灌注小鼠学习记忆能力的影响及其作用机制。方法:昆明种小鼠70只随机分成5组,分别为假手术组,模型组,穿山龙总皂苷高、中、低剂量组(100、50、25 mg·kg-1·d-1),灌胃给药连续15 d。给药12 d采用双侧颈总动脉夹闭,建立反复脑缺血再灌注致小鼠学习记忆障碍模型,术后24 h和48 h通过跳台、Y型电迷宫实验,观察穿山龙总皂苷对反复脑缺血再灌注小鼠学习记忆能力的影响,同时对脑组织中超氧化物歧化酶(SOD)活性和丙二醛(MDA)、一氧化氮(NO)含量进行检测。结果:与模型组比较,穿山龙总皂苷可明显改善反复脑缺血再灌注小鼠的学习记忆能力(P<0.05或P<0.01),增强脑组织中SOD活性(P<0.05或P<0.01),降低MDA和NO含量(P<0.05或P<0.01)。结论:穿山龙总皂苷对反复脑缺血再灌注损伤小鼠的学习记忆功能具有改善作用,其机制可能与增强脑组织中SOD活力,降低MDA和NO含量有关。
Objective : To observe the effects and mechanism of the total saponins from Dioscorea Nipponica Makino on studying and memory in mice with repeatedly cerebral ischemia reperfusion. Methods :70 Kunming mice were randomly di- vided into five groups, sham - operation group, model group, the total saponins from Dioscorea Nipponica Makino groups ( 100,50,25 mg · kg- 1· d- 1 respectively), mice in treatment groups were intragastrically administrated with different do- ses of the total saponins from Dioscorea Nipponica Makino for consecutive 15 days. After 12 days,mice model of cerebral ischemia - reperfusion injury was established by bilateral common carotid artery blocking. After 24 and 48 hours, behavior of learning and memory was observed by step down test and electric maze test to observe the effects of the total saponins from Dioscorea Nipponica Makino on studying and memory in mice with repeatedly cerebral ischemia reperfusion. The con- centration of malondialdehyde (MDA) and nitric oxide(NO) while activity of superoxide dismutase(SOD) in brain tissue was detected. Results : Compared with model group, the total saponins from Dioscorea Nipponica Makino obviously improved the disorder of studying and memory of mice (P 〈 0.05 or P 〈 0.01 ) , and inhibited the abnormal increased MDA and NO ( P 〈 0.05 or P 〈 0.01 ) while increased activity of SOD (P 〈 0.05 or P 〈 0.01 ) in brain tissue caused by repeated cer- ebral ischemia reperfusion. Conclution :The total saponins from Dioscorea Nipponica Makino can protect mice being repeat- edly cerebral ischemia reperfusion from the damage of studying and memory, and its mechanism is related to increasing SOD activity and lowering MDA and NO content in brain.
