摘要
空育131和稻花香是黑龙江省很有代表性的寒地栽培品种,也是育种研究的重要材料。为了获取此品种的优质RNA,以经低温避光处理3d的空育131及稻花香水稻的10日龄水稻苗叶为材料,利用优化的UNIQ-10柱式Total RNA Extractor(Trizol)试剂盒法提取RNA。之后,进行RT-PCR反应,并对RT-PCR的结果进行琼脂糖凝胶电泳的检测。结果表明:通过优化,改进了UNIQ-10柱式Total RNA Extractor(Trizol)试剂盒法,提取的RNA纯度较高,A260/A280均接近1.8,并且RT-PCR的条带清晰。
Kongyul31 and Daohuaxiang are typical cultivars in Heilongjiang province. And they are important materials of breeding research. In order to obtain their high-quality RNA, Kongyu131 and Daohuaxiang rice seedling leaves were used as materials, and treated with low temperature away from light for three days, whose age were ten days. Optimization of UNIQ-10 pillar TotalRNA Extractor (Trizol) kits was used to extract RNA. After RT-PCR reaction.and the results of RT-PCR by detection of agarose gel electrophoresis. The result showed that by the optimization,the UNIQ-10 pillar TotalRNA Extractor (Trizol) kits could obtain RNA with high purity,whose A260/A280 was close to 1.8,and stripe of RT-PCR was clear.
