响应面优化西兰花中萝卜硫素的超声辅助提取工艺 被引量:10

Optimization on extraction techniques of sulforaphane from broccoli with ultrasound-assisted by response surface methodology

摘要 对超声辅助提取西兰花中萝卜硫素的工艺进行了研究,以西兰花鲜食部分绿色幼嫩花茎和花蕾为试验材料,利用单因素及响应面法对外源芥子酶酶解西兰花后,超声波辅助乙醇提取萝卜硫素的工艺参数进行筛选及优化,在单因素试验基础上选取酶解时间,料液比和提取时间3个因素,再根据Central Composite中心复合试验设计原理采用三因素三水平的响应面分析法,确定各因素对萝卜硫素提取率的影响程度.结果表明:超声辅提取西兰花中萝卜硫素的最佳提取条件为:酶解时间0.95h,料液比1∶41.8,提取时间1.1h,理论提取率为1 890.81μg/g,验证提取率为1 814.59μg/g,其相对误差为4.20%,表明该提取工艺参数在西兰花中提取萝卜硫素是可行的. Fresh parts of broccoli were chosen as experimental material,and extraction techniques were optimized on sulforaphane from broccoli with the ultrasound-assisted.The process parameters of extracted by ethanol with assistance of ultrasonic wave were screened and optimized though the method of single factor and response surface after exogenous myrosinase hydrolyze the fresh part.Enzymolysis time,solid-liquid ratio and extraction time were screened on the basis of single factor experiment,the effects of various factors on extraction ratio were determined by the method of response surface analysis with three factors and three levels according to the central composite design method of Central Composite.The optimum extraction conditions were as enzymolysis time 0.95 h,solid-liquid ratio 1∶41.8,extraction time 1.1h.Thetheoretical extraction rate reached 1 890.81μg/g and the verification extraction rate was 1 814.59μg/g,and the relative error was 4.20%.The results showed that these parameters were practicable to extract sulforaphane from broccoli.
