固本止咳中药对慢性阻塞性肺疾病模型小鼠肺组织中弹性蛋白酶表达的影响 被引量:8

Effects of Gubenzhike Recipe on expression of neutrophil elastase in the lung tissue of COPD mice

摘要 目的:观察固本止咳中药对慢性阻塞性肺疾病(COPD)模型小鼠肺组织中中性粒细胞弹性蛋白酶(NE)表达的影响,探讨固本止咳中药治疗COPD抗炎性反应的作用机制及NE与分泌型免疫球蛋白A(SIg A)的内在联系。方法:采用鼻腔滴入脂多糖加熏香烟的方法建立COPD小鼠模型,分为空白对照组、模型对照组、固本止咳中药组;运用免疫组化方法检测小鼠肺组织中NE表达水平,采用ELISA方法检测小鼠气道SIg A分泌水平。结果:模型对照组肺组织中NE表达气道SIg A分泌水平较空白对照组显著增多(P<0.01,P<0.05),固本止咳中药组较模型对照组显著减少(P<0.01,P<0.05)。结论:固本止咳中药可改善COPD模型小鼠肺组织及气道黏膜炎性浸润状态。 Objective: To observe the effects of Gubenzhike Recipe(GR) on the expression of neutrophil elastase(NE) in lung tissue of chronic obstructive pulmonary disease(COPD) mice, and to explore its mechanism on anti-inflammatory response and the internal relations of NE and secretory immunoglobulin A(SIg A). Methods: The COPD model was established by exposure to cigarette smoke in combination with intra-nostrils injection of lipopolysaccharide(LPS), and divided into control group, model group and GR group. The expression of NE in lung tissue was tested by immunohistochemistry, while the SIg A level in respiratory tract was detected by method of ELISA. Results: The expression of NE in lung tissue and in the model group was increased than that in the control group(P〈0.01, P〈0.05), while in the GR group was decreased than that in the model group(P〈0.01, P〈0.05). Conclusion: Gubenzhike Recipe could alleviate the inflammatory infiltration state of the COPD mice' airway mucosa and lung tissue.
