卷柏各部位对糖尿病大鼠胰岛组织形态及骨骼肌葡萄糖转运蛋白-4表达的影响 被引量:7

Effects of different fractions from Selaginella Tamariscina(Beauv.) Spring on histomorphology of pancreas and the expression of GLUT-4 protein in skeletal muscle in diabetic rats

摘要 目的:观察卷柏醇提物所得各部位对糖尿病大鼠胰岛组织形态及骨骼肌葡萄糖转运蛋白-4(GLUT-4)表达的影响。方法:通过腹腔注射链脲佐菌素(STZ,65mg/kg)诱导糖尿病大鼠模型,观察卷柏各部位给药7d后,空腹血糖、餐后4h血糖变化,观察胰岛组织HE染色切片,并通过免疫组化的方法观察GLUT-4的表达情况。结果:卷柏80%乙醇部位组大鼠空腹血糖及餐后4h血糖与模型组比较均显著降低(P<0.05),GLUT-4的表达量比模型组显著增加(P<0.01),胰岛组织形态与模型组比较也有所改善。结论:卷柏80%乙醇部位可能是卷柏降糖的活性部位,其降糖作用机制可能与修复受损的胰岛组织,增强骨骼肌GLUT-4表达,进而增强机体胰岛素敏感性有关。 Objective: To study the effects of different fractions from Selaginella Tamariscina(Beauv.) Spring on histomorphology of pancreas and the expression of glucose transport type-4(GLUT-4) protein in skeletal muscle in diabetic rats. Methods: The diabetic rat models were induced by intraperitoneal injection of STZ(65mg/kg), and treated with the different fractions from Selaginella Tamariscina(Beauv.) Spring for 7d. The serum levels of fasting blood glucose(FBG) and 4h postprandial blood glucose(4h PG) were measured, meanwhile pathologic-morphologic of pancreas was observed by HE staining, the skeletal muscle GLUT-4 expression was detected by immunohistochemical staining. Results: Compared with the model group, the 80% ethanol fraction could decrease the FBG and 4h PG(P〈0.05), and increase the skeletal muscle GLUT-4 protein expression(P〈0.01), meanwhile improve islet tissue repair function. Conclusion: The results suggest that the 80% ethanol fraction is the activity fraction of Selaginella Tamariscina(Beauv.) Spring. The inhibitory effect of active of the Selaginella Tamariscina(Beauv.) Spring on diabetes might be associsted with promoting the skeletal muscle GLUT-4 protein expression and restore the pancreas to enhance insulin sensitivity.
