摘要
目的:观察清热利胆解毒方对铜负荷大鼠学习记忆和海马区超微结构的影响。方法:100只雄性大鼠分为空白(NG)组、模型(MG)组、清热利胆解毒方(TCM)组、青霉胺(PCA)组和清热利胆解毒方加青霉胺(TCM+PCA)组。Morris水迷宫实验测试大鼠学习记忆能力,电镜观察大鼠海马超微结构变化。结果:定位航行试验中MG组大鼠的平均潜伏期较NG组明显延长(P<0.01),各治疗组较MG组平均潜伏期显著减少(P<0.01)。空间探索试验中MG组大鼠的穿越原平台位置次数较NG组明显减少(P<0.01),各治疗组较MG组显著增加(P<0.01)。与NG组比较,MG组大鼠神经元突触间隙厚度明显变宽(P<0.05),PSD厚度明显变薄(P<0.05)。各治疗组较MG组突触间隙明显变窄(P<0.05),PSD厚度明显增加(P<0.05)。结论:清热利胆解毒方可能通过调节神经元突触界面超微结构来发挥改善学习记忆功能的作用。
Objective: To observe effects of Qingre Lidan Jiedu Formula(QLJF) on learning and memory and hippocampal ultramicro-structure of copper-load rat model. Methods: One hundred male rats were randomly divided into anormal control(NG) group, model control(MG) group, Chinese medicine(TCM) group, penieillamine(PCA) group and TCM+PCA group. The learning and memory behavior were tested by Morris water maze. The hippocampal ultramicro-structure as observed by transmission electron microscope. Results: In place navigation test, the mean latency of in the MG group obviously prolonged and the number of traversing flat roof obviously decreased than that of the NG group(P〈0.01), while of each treatment group significantly increased compared with that of the model group(P〈0.01). Compared with the NG group, the synaptic cleft was obviously broaden in the MG group(P〈0.05), the PSD thickness decreased(P〈0.05). The synaptic cleft in each treatment group significantly narrowed compared with the MG group(P〈0.05), and the PSD thickness significantly increased(P〈0.05). Conclusion: QLJF might play a role in improving learning and memory function through adjusting the neuron ultrastructure.
