健脾化瘀方对肝癌耐药细胞P-糖蛋白的影响

Effects of Jianpi Huayu Formula on drug resistance cell P-glycoprotein in liver cancer

目的:研究健脾化瘀方对肝癌耐药细胞bel-7402/5-FU表面耐药蛋白P-糖蛋白(P-gp)表达的影响,探寻健脾化瘀方可能的逆转耐药的机制。方法:选择肝癌细胞bel-7402、肝癌耐药细胞bel-7402/5-FU做为实验细胞,并建立BALB/c裸鼠肝癌耐药细胞皮下移植瘤模型,运用PCR及western blot法检测健脾化瘀方给药后P-gp蛋白的表达水平。结果:经健脾化瘀方给药后,bel-7402/5-FU细胞P-gp的表达水平下降;中药组瘤体质量小于bel-7402组;中药组瘤体中P-gp的表达水平下降。结论:健脾化瘀方可以降低耐药细胞中P-gp蛋白的表达水平,并可能是通过该途径逆转细胞的耐药性。

Objective： To explore the effects of Jianpi Huayu Formula（JHF） on the expression of P-glycoprotein（P-gp） in bel-7402/5-FU liver cancer cell, and the possible reversal mechanism. Methods： The bel-7402 cell of liver cancer cell and bel-7402/5-FU cell of liver cancer resistant cell were selected as experimental cells, and subcutaneous tumor model of drug resistance liver cancer cell was established with BALB/c nude mice. The expression level of P-gp was detected with PCR and western blot after the treatment of JHF. Results： After the treatment of JHF, the expression level of P-gp in bel-7402/5-FU cell was decreased; JHF could inhibit the growth of tumor in the Chinese medicine group; the expression level of P-gp in tumors was decreased in the Chinese medicine group. Conclusion： JHF could decrease the expression level of P-gp, and it might reverse drug resistance in this way.
