槐花散总黄酮大孔树脂分离纯化的工艺研究 被引量:10

摘要 目的:研究大孔树脂分离纯化槐花散总黄酮的的工艺条件和参数。方法:采用紫外分光光度法测定总黄酮含量,选择了3种大孔树脂和1种聚酰胺树脂,以槐花散总黄酮吸附率和解吸率为指标,筛选出较优的树脂,并对分离纯化的主要影响因素进行考查,优化工艺条件。结果:AB-8大孔树脂对槐花散总黄酮有良好的纯化性能,适宜的工艺条件为:上样药液浓度为1.5mg/m L,上样体积为2BV,先用4BV水洗,再用3BV 65%乙醇洗脱,树脂径高比为1∶9。结论:AB-8大孔树脂分离纯化槐花散总黄酮效果好、工艺稳定。 Objective: To study the technological condition and parameters for separation and purification of total flavonoids from Huaihua San. Methods: The content of total flavonoids was analyzed by ultraviolet spectrophotometry. Three kinds of macroporous resins and one kind of polyamide resins were valuated by the adsorption and desorption ratios to choose the suitable resin. The main process conditions were ascertained to optimize the process parameters. Results: AB-8 resin showed advantageous adsorption and desorption properties. The optimal conditions were determined as follows: the sample concentration was 1.5g/m L and its volum was 2BV, the sample was eluted first with 4BV of water, then with 3BV 65% of ethanol, the ratio of diameter to height of resin was 1∶9. Conclusion: AB-8 resin method has a good effect and is stabile for separation and purification of total flavonolds from Huaihua San.
