摘要
目的探讨蛇葡萄素(AMP)对人肾癌786-O细胞株增殖与凋亡的作用。方法将不同浓度的AMP作用于体外培养的肾癌786-O细胞株,采用MTT法检测不同浓度的AMP对肾癌786-O细胞增殖的作用的影响;采用Annexin V/PI双染法检测不同浓度的AMP对肾癌786-O细胞凋亡的作用。结果 MTT结果显示AMP能显著抑制肾癌786-O细胞增殖,半数抑制浓度(IC50)为17.23μg/mL;经不同浓度的AMP作用7h后,肾癌786-O细胞凋亡率明显增加,AMP浓度为10μg/mL和20μg/mL作用于肾癌786-O细胞后,细胞凋亡率升高(P<0.05);而AMP浓度为40μg/mL和80μg/mL作用细胞后,细胞凋亡率显著升高(P<0.01)。结论 AMP在体外能显著抑制肾癌786-O细胞增殖,且能促进肾癌786-O细胞凋亡。
Objective To explore the effects of ampelopsin on proliferation and apoptosis of human renal cancer cell line 786- O. Methods Different concentrations of ampelopsin were used on the renal cancer cell lines 786-O,MTT method was used to detect the effects of different concentrations of ampelopsin on cell proliferation of renal cancer cell 786-O; Annexin V/PI double staining was used to detect the effects of different concentrations of ampelopsin on cell apoptosis of renal cancer cell 786-O. Results The MTT results showed that the ampelopsin could significantly inhibit renal cancer cell proliferation,and ICS0 was 17.23 /zg/mL. The apoptosis rates of renal cancer cell 786-O increased significantly after 7 h with different concentrations of ampelopsin;the cell apop- tosis rates of ampelopsin of 10 μg/mL and 20 μg/mL of renal cancer 786-O cell were increased (P〈0.05) ;the cell apoptosis rate of concentrations of 40 μg/mL and 80 μg/mL increased significantly (P〈0.01). Conclusion Ampelopsin can significantly inhibit the cell proliferation of renal cancer cell 786-O,and can promote renal cancer cell 786-0 apoptosis.
