湿疹不同证候核因子-κB、热休克蛋白70的表达 被引量:2

Expression of NF-κB and Heat Shock Protein 70 in skin lesion of patients with eczema in different syndromes

摘要 目的:观察湿疹不同证候核因子-κB、热休克蛋白70表达,探讨其与湿疹不同证候发生的关系。方法:收集湿疹患者30例,包括风湿热蕴证18例,血虚风燥证12例,另取健康人8名作为对照。荧光定量PCR检测各组组织中NF-κB mRNA和HSP70mRNA表达水平,Western blot法检测皮损内核因子-κB、热休克蛋白70蛋白表达。结果:HSP70 mRNA和蛋白表达,风湿热蕴证组高于血虚风燥证组和健康对照组,差异有统计学意义;血虚风燥证组表达与健康对照组比较差异无统计学意义。NF-κB mRNA和蛋白表达,风湿热蕴证组表达高于血虚风燥证组和健康对照组,差异有统计学意义;血虚风燥证组表达高于健康对照组,差异有统计学意义。结论:HSP70和NF-κB的表达程度可能体现了"湿疮"病理过程中机体正邪力量,为揭示中医湿疮"证候"的本质提供了理论依据。 Objective: To investigate the expression of Heat Shock Protein 70( HSP70) and NF-κB p65 in skin lesion of patients with eczema in different syndromes and the roles of them in eczema of different syndromes. Methods: 30 cases of patients with eczema were collected,18 cases were wind-damp-heat syndrome,12 cases were blood deficiency and wind dryness syndrome,meanwhile,8 of healthy people were as control. The mRNA expression of NF-κB and HSP70 in tissues were determined quantitatively with FQ-PCR. The protein expression of NF-κB and HSP70 in tissues were determined quantitatively with western blot. Results: The mRNA and protein expression of HSP70 in the wind-damp-heat syndrome group was higher significantly than that in the blood deficiency and wind dryness syndrome group and the control group.While there was no significantly different between the blood deficiency and wind dryness syndrome group and the control group. The mRNA and protein expression of NF-κB p65 in the wind-damp-heat syndrome group was higher significantly than that in the blood deficiency and wind dryness syndrome group and the control group; The mRNA and protein expression of NF-κB p65 in the blood deficiency and wind dryness syndrome group was higher significantly than that in and the control group. Conclusion: The expression level of NF-κB p65 and HSP70 may reflect the strengths of pathogens and vital Qi respectively. The result provided some clues to investigate the Chinese medicine "syndrome"nature of eczema.
