摘要
目的:观察帕金森病(PD)模型大鼠黑质致密部(SNc)免疫球蛋白G(IgG)及其mRNA的表达变化,探讨Ig G在PD发病中的作用。方法:单侧微量注射6-羟多巴胺(6-OHDA)制备PD大鼠模型。免疫组化法观察对照组大鼠双侧、模型组大鼠健侧及注射侧SNc酪氨酸羟化酶(TH)阳性神经元和IgG阳性细胞的数量变化,RTPCR法检测两组大鼠黑质TH mRNA、IgG mRNA的表达变化。结果:模型组大鼠注射侧SNc内TH阳性神经元的数量较健侧显著减少(P<0.05),两组大鼠SNc均可见IgG阳性细胞,但模型组大鼠注射侧SNc IgG阳性细胞的数量显著增加(P<0.05),染色较深。模型组大鼠注射侧TH mRNA表达明显低于健侧和对照组双侧,IgG mRNA表达明显高于健侧和对照组双侧。结论:PD大鼠黑质致密部细胞IgG蛋白及其mRNA表达均增加,表明IgG与PD的发生有关。
Objective: To investigate the expression changes of immunoglobulin G( IgG) and IgG mRNA in substantia nigra compacta par( SNc) in rat model of Parkinson's disease( PD) in order to explore the role of IgG in PD. Methods:PD rat model was established by microinjection of 6-hydroxydopamine( 6-OHDA) solution into the substantia nigra of the right side of the male rat brain. The numerical changes of tyrosine hydoxylase( TH)-and Ig G-positive cells on two sides of rats in control and model groups were examined by immunohistochemical method. The expression changes of TH and Ig G at the mRNA level in substantia nigra were detected by RT-PCR. Results: The number of TH-positive cells on the 6-OHDA-injected side of the substantia nigra in the model rats was significantly reduced( P〈0. 05). Both of the two groups were observed to have many IgG-positive cells in SNc,however,there were more Ig G-positive cells on the 6-OHDA-injected side of the model group( P〈0. 05). The expression of TH mRNA on the injected side in SNc of the model group was reduced,while the expression of Ig G mRNA was increased significantly compared with those of the control group. Conclusion: The expression of Ig G and Ig G mRNA were significantly increased in PD rat models. suggesting that Ig G is associated with PD.
