一种昆明种属来源的视网膜变性小鼠同源导入C57BL/6J小鼠近交系的表型研究

摘要 目的将本实验室发现的一种昆明种属来源的自发性遗传性视网膜变性小鼠(暂命名为KMrdf/rdf小鼠)的致病基因同源导入C57BL/6J小鼠,培育一种新的近交系小鼠(暂命名为B6rdf/rdf小鼠),并对B6rdf/rdf小鼠进行眼部表型的发育学研究。方法采用眼底照相、眼底荧光素血管造影、组织形态学和视网膜电图观测不同年龄B6rdf/rdf小鼠的眼部表型。结果眼底照相和荧光素血管造影发现B6rdf/rdf小鼠表现为进行性视网膜血管退化及脱色素改变,视网膜组织形态学表现为外核层在P14显著变薄,外核层和外网状层至P21已基本消失,视网膜电图自P14已记录不到明显波形。结论 B6rdf/rdf小鼠眼部表型呈快速进行性改变,与KMrdf/rdf小鼠表型相似。 Objective To cross spontaneously hereditary retinal degeneration mice( temporarily designated as KMrdf / rdfmice) derived from Kunming mice with C57 BL /6J mice to establish a congenic inbred strain( temporarily designated as the mice),and study the ocular phenotype of the mice. Methods Fundus photography, fundus fluorescein angiography, tissue morphology and electroretinography( ERG) were performed to observe the ocular phenotype of B6 rdf / rdfmice at different ages. Results The fundus photography and fundus fluorescein angiography showed progressive retinal vascular degeneration and depigmentation. The tissue morphology showed that the outer nuclear layer was significantly thinned at P14,and the outer nuclear layer and outer plexiform layer almost disappeared at 21 d post born. No obvious waveform was observed from14 d after born in the ERG. Conclusion The ocular phenotype of B6 rdf / rdfmice was similar to that of KMrdf / rdf mice,which indicates that B6 rdf / rdfmice can be used as an retinitis pigmentosa mouse model.
