环磷酸腺苷活化交换蛋白对心肌重塑中Ca^(2+)循环的调控作用 被引量:3

The regulation role of Epac for calcium circulation in myocardial remodeling

摘要 心肌重塑是心肌细胞应对不良心血管因素刺激做出的代偿性反应,重塑过程中涉及心肌细胞内外Ca2+循环的平衡,作为鸟苷酸交换因子的环磷酸腺苷活化交换蛋白(exchange proteins directly activated by cAMP,Epac)对维持这种平衡起着重要的调控作用。在心力衰竭患者因Epac表达上调,使其对心肌细胞内外Ca2+循环平衡调控出现异常而致心肌泵衰竭,进而导致患者死亡,明确Epac对心肌重塑中Ca2+循环的调控作用对于研发治疗心力衰竭新的靶向药物具有重要意义。 Myocardial remodeling is a mechanism by which the cardiomyocytes responds to adverse stimulation induced by cardiovascular factors in an effort to make a compensatory response.The remodeling process relates to the calcium homeostasis inside and outside of the cardiomyocytes.Epac,which belongs to a family of guanine nucleotide exchange factors,plays a crucial role in maintaining this homeostasis.The cardic pump is dysfunctional when the calcium homeostasis inside and outside of the cardiomyocytes is disorder,which is caused by the up regulation of Epac expression in patients with heart failure.This ulmately lead to the patients’ death.It is important to illustrate the mechanisms for Epac regulating the calcium circulation in myocardial remodeling and develop new targeted therapy drugs for patients who suffer from heart failure.
