血吸虫虫卵抗原对巨噬细胞和肺动脉内皮细胞IL-1β表达的影响 被引量:1

摘要 目的探讨血吸虫虫卵抗原(SEA)对巨噬细胞RAW264.7和肺动脉内皮细胞(HPAECs)上清液中白细胞介素1β(IL-1β)表达的影响。方法体外培养RAW264.7细胞和HPAECs,随机分为对照组(A组)、脂多糖+SEA组(B组)和SEA组(C组),采用ELISA法检测细胞上清液中IL-1β表达量。结果与A组相比,B组和C组细胞上清液中IL-1β的表达均上调(P<0.05);C组IL-1β的表达低于B组(P<0.05)。结论 SEA诱导巨噬细胞和HPAECs上清液中IL-1β的表达,提示SEA诱导的炎症可能参与血吸虫病相关性肺动脉高压的发生。 Objective To investigate the effect of schistosoma egg antigen (SEA ) on IL‐1βexpression in the supernatant of macrophage RAW264 .7 and human pulmonary artery endothelial cells (HPAECs) .Methods RAW264 .7 cells and HPAECs were cultured in vitro and randomly divided into three groups of A(normal control) ,B(induced with LPS 2.5 μg/ml followed by SEA 50 μg/ml after 3 h) and C (induced with SEA 50 μg/ml) .IL‐1β expression in the supernatant of cells was detected by ELISA .Results Compared with group A ,IL‐1β expression in the supernatant of RAW264 .7 cells and HPAECs was up‐regulated in groups of B and C(P〈0 .05) .IL‐1βexpression in group C was lower than that in group B(P〈0 .05) .Conclusion SEA promotes IL‐1β expression in RAW264 .7 cells and HPAECs ,which indicates that SEA‐induced inflammation may participate in the pathogenesis of schistosomiasis‐associated pulmonary arterial hypertension .
