白细胞介素-17基因多态性与侵袭性牙周炎的相关性研究 被引量:8

摘要 目的已有少量文献证实白细胞介素-17(interleukin-17,IL-17)基因多态性与多种免疫炎症性疾病相关。本研究旨在探索中国汉族人群中,IL-17A(rs2275913)和IL-17F(rs763780)位点单核苷酸多态性(single nucleotide polymorphisms,SNPs)与侵袭性牙周炎(aggressive periodontitis,Ag P)的相关性。方法采集87例Ag P、79例牙周健康者的外周静脉血,提取基因组DNA,采用多重SNa Pshot技术分析待测位点的基因多态性。结果 IL-17A(rs2275913)位点的基因型分布、IL-17F(rs763780)位点的基因型和等位基因分布在Ag P组和健康对照(healthy controls,C)组中差异均无统计学意义(P≥0.05)。IL-17A(rs2275913)位点在Ag P组中A等位基因频率显著高于健康对照组(54.0%vs.42.4%,P=0.034,OR=1.596,95%CI=1.034-2.463)。结论研究结果显示IL-17A(rs2275913)位点A等位基因可能是Ag P发病的危险因素,提示IL-17A(rs2275913)位点的多态性可能与中国汉族人群的Ag P易感性有关。 Objective Interleukin-17 (IL-17) polymorphisms have been testified by a few studies to be associated with several im- mune inflammatory diseases. The aim of present study was to explore whether the single nucleotide polymorphisms (SNPs) in IL-17A (rs2275913) and IL-17F (rs763780) genes were correlated with the susceptibility to aggressive periodontitis (AgP) in Chinese Han population. Methods 87 AgP patients and 79 healthy controls were recruited in this study. Genomic DNA was extracted from the pe- ripheral blood samples,and gene polymorphisms were determined by multiplex SNaPshot technique. Results No significant differences were observed in genotype or allele frequency distributions of IL-17A (rs2275913) and IL-17F (rs763780) between AgP and healthy controls (P i〉 0, 05 ). However, the A allele frequencies of IL-17 A (rs2275913 ) were significantly higher in AgP patients compared with healthy controls (54.0% vs.42.4% ,P=0.034,OR=1.596,95%CI=1. 034-2. 463 ). Conclusions These findings indicate that the IL-17A (rs2275913) polymorphism seemed to be a genetic risk factor of AgP. It suggests that the IL-17A( rs2275913 ) polymorphism may be associated with the susceptibility to AgP in Chinese Han population.
