摘要
目的研究银翘散对铜绿假单胞菌生物膜的影响及与阿奇霉素的协同作用。方法选取铜绿假单胞菌野生株POA1建立体外生物膜模型;微量倍比稀释法检测银翘散、阿奇霉素对铜绿假单胞菌的最低抑菌浓度;棋盘稀释法测定银翘散和阿奇霉素的协同抗菌作用;银染法观察药物对生物膜形态的影响。结果银翘散抑制铜绿假单胞菌生物膜形成的MIC是500g/L,阿奇霉素抑制铜绿假单胞菌生物膜形成的MIC是250.0mg/L,两药联合后MIC分别为125.0g/L和62.5mg/L,抑菌分级指数是(FIC)是0.5,提示两药协同作用明显。结论银翘散具有抗铜绿假单胞菌生物膜作用,与阿奇霉素有协同抗菌作用。
Objective To investigate the effects of Yinqiao powder against Pseudomonas aeruginosa biofilms and its synergistic with azithromycin. Methods The wild type PAO1 of Pseudomonas aeruginosa were cultured to establish biofilm formation.Biofilm was observed after AgNO3 staining. Minimum inhibition concentrations (MICs) of Yinqiao powder and azithromycin were measured by doubling dilution. Checkerboard method was used to examine synergism of Yinqiao powder and azithromyein. Results MIC of Yinqiao powder against Pseudomonas aeruginosa biofilms was 500.0g/L and azithromyein 250.0mg/L. After the combination of two-drugs the MIC was 125.0g/L and 62.5mg/L. Fractional inhibitory concentration (FIC) was 0.5,which showed a synergism with azithromycin. Conclusion Yinqiao powder could inhibit the formation of Pseudomonas aeruginosa biofilms,and there is a synergistic effect in combination with Azithromycin.
