健脾法对肠道甜味觉信号分子mRNA表达的影响 被引量:2

摘要 目的研究健脾方案对肠粘膜上皮甜味觉通路中味觉受体细胞及信号关键分子mRNA表达的影响。方法将功能性腹泻患者分为脾胃虚弱组及脾胃湿热组(各20例),脾胃虚弱组服用健脾I号方,脾胃湿热组服用健脾II号方,每次20m L,3次/d,疗程4周。胃镜下钳取治疗前后十二指肠粘膜组织2块,采用QPCR法检测两组功能性腹泻患者治疗前后十二指肠粘膜味觉通路中味觉受体第一家族(T1R1、T1R2、T1R3)及信号关键分子的表达。结果治疗后除脾胃湿热组肠道味觉受体T1R1外,两组肠道味觉受体(T1R1、T1R2、T1R3)及关键信号分子Gαgust表达均有升高(P<0.01)。治疗后,脾胃虚弱组肠道味觉受体第一家族及信号关键分子表达高于经健脾II号方治疗后的脾胃湿热组(P<0.01)。结论健脾法可有效增加功能性腹泻患者肠道甜味觉受体细胞及关键信号分子mRNA的表达。 Objective To study the effect of spleen strengthening method on expression of gustatory receptors and key signal molecules in intestinal tracts of cases with functional diarrhea. Methods Patients with functional diarrhea were divided into spleen-stomach deficiency group comprised of 20 cases and treated with prescription I and spleen-stomach damp- heat group comprised of 20 cases and treated with prescription II ,20mL each time and 3 times per day for 4 weeks.Duodenal mucosal tissues from patients were obtained and QPCR was used to measure the expression level of gustatory receptors and key signal molecule Gotgust before and after treatment. Results After treatment, expressions of gustatory receptor (T1R1,T1R2, T1R3) and Getgust were both increased in patients of the two groups (P〈O.O1) except the T1R1 in spleen-stomach damp-heat group. The expressions of T1R1, T1R2, T1R3, Gotgust in patients of spleen-stomach deficiency group were higher than that of spleen-stomach damp-heat group (P〈0.01) Conclusions The spleen strengthening method can influence the expression mRNA of taste receptors and key signal molecules in intestinal tracts of cases with functional diarrhea.
