摘要
目的:研究藻黄合剂防治糖尿病肾病的作用机制。方法:体外培养人肾小球系膜细胞并在高糖培养基中孵育,用不同浓度的藻黄合剂培养液孵育高糖环境下培养的GMC,于不同时间(24h、48h)采用MTS方法检测各组系膜细胞增殖的变化。分别观察24h、48h系膜细胞增殖和FN的分泌。结果:不同浓度的ZHM作用于GMC 24h、48h,除低剂量组24h无显著作用外(P>0.05),其余各组均能显著抑制GMC的增殖(P<0.01或P<0.05)。藻黄合剂各剂量组在24h、48h时均能显著抑制高糖下FN的分泌(P<0.01或P<0.05)。结论:ZHM在一定时间内可明显抑制体外培养的GMC在高糖环境下的增殖,同时抑制FN的分泌,这可能是藻黄合剂延缓糖尿病肾病、保护肾功能的作用机制之一。
Objective: To investigate mechanism of preventing and treating diabetic nephropathy with the ZaoHuang mixture. Methods: GMCs were brought up in vitro and hatched in high glucose medium. Different times of GMC (24 h, 48 h), hatched in high glucose medium, were brought up in different degrees of the Zaohuang mixture. Groups of GMCs in proliferation of change were tested by MTS. GMCs in proliferation of change and Secretion of FN was observed at 24h, 48h. Results: According to different concentrations of ZHM in GMC24h, 48h, except the low-dose group 24h had no significant effects (P>0.05). The rest of the groups could significantly inhibit the proliferation of GMC’s (P<0.01 orP<0.05). Each dose group of the Zaohuang mixture in 24h, 48h significantly inhibited the secretion of FN under high glucose environment (P<0.01 orP<0.05). Conclusion: ZHM can inhibit the vitro proliferation of GMC in high glucose environment within a certain time, while inhibiting the secretion of FN, which may be one of the mechanisms of the Zaohuang mixture in delaying diabetic nephropathy and protecting renal function.
