口腔科手机润滑油局部注射小鼠致炎性研究

Proinflammatory study of dental lubricating oil injected locally in mice

目的研究口腔科手机润滑油局部注射小鼠的致炎情况。方法取口腔科手机润滑油原液及5.66 mg、2.92 mg、2.02 mg润滑油和生理盐水的混合液各0.1 m L,分别对4组(每组6只)小鼠进行皮内注射。对照组注射无菌生理盐水0.1 m L,1次/d,连续5 d,在同一部位作皮内注射,观察其皮肤局部反应,第6天全部处死,光学显微镜下观察其组织病理变化。结果小鼠局部注射溶液后,注射皮丘出现不同程度的消退,实验组注射部位出现微红或红肿,无糜烂等表现,对照组注射部位无明显红肿。病理组织学检查,实验组注射部位真皮内可见圆形或卵圆形空腔,空腔的间隔和其周围可见纤维组织增生及巨噬细胞、中性白细胞等炎症细胞浸润,且随着润滑油含量的增加,炎症细胞浸润程度趋于严重;对照组组织学结果显示,注射部位真皮内未见圆形或卵圆形空腔,见少量纤维组织增生,无明显炎症细胞浸润。5组小鼠炎症反应程度差异有统计学意义(χ2=33.065,P<0.001)。结论口腔科手机润滑油可引起小鼠局部炎症反应,其程度与润滑油含量有关。

Objective To detect the inflammatory effect of dental lubricating oil injected locally in mice .Methods Four experimental groups of mice received daily intradermal injection with 0.1 ml pure dental lubricating oil or saline-mixture of 5.66 mg, 2.92 mg, 2.02 mg dental lubricating oil separately for 5 d.The group only injected with normal sa-line was treated as control.The local skin reactions were observed.The mice were sacrificed at the sixth day and the his-topathologic changes were examined under microscope.Results After local injection, injection pichel appeared different degree of fading, the experimental group at the injection site appears reddish or slightly redness with no erosion perform-ance, the control group illustrated no obvious swelling at the injection site.The histologic examination showed round or o-void cavities in dermis at the injection site in the experimental groups and the cavities were surrounded by hyperplastic fi-brous tissue with infiltration of macrophages, neutrophils, and other inflammatory cells.With the increase of lubricating oil dose, the infiltration of inflammatory cells became more serious.There was no such inflammatory response except for little fibrous hyperplasia in the control group.Difference of five groups of mice inflammation degree was statistically sig-nificant （χ2 =33.065, P〈0.001）.Conclusion Dental lubricating oil may cause local inflammatory response in mice, and the degree of inflammationare associated with the amount of oil.
