人永生化角质形成细胞急性光损伤程度与线粒体膜电位变化的关系

摘要 目的:探讨人永生化角质形成细胞(Ha Ca T细胞)急性光损伤程度与线粒体膜电位变化的关系。方法:用不同剂量的UVB照射Ha Ca T细胞,通过光学显微镜观察细胞形态学改变,MTT检测细胞增殖活性(吸光度),JC-1荧光染料法检测线粒体膜电位的改变。结果:100 m J/cm2剂量UVB照射可引起Ha Ca T细胞形态发生变化,与空白对照组相比增殖活性降低(P<0.05),线粒体红色/绿色荧光强度值明显低于正常对照组(P<0.05);随着照射剂量增加,红色/绿色荧光强度值逐渐下降。结论:线粒体膜电位的降低可以反映Ha Ca T细胞急性光损伤的程度,线粒体膜电位越低,Ha Ca T细胞急性光损伤的程度越大。
