HPLC测定葛根温胆颗粒中葛根素的含量

Content Determination of Puerarin in Gegen Wendan Granules by HPLC

目的：建立葛根温胆颗粒中葛根素的含量测定方法。方法：采用高效液相色谱法,Prontosil 120-5-C18柱（250 mm×4.6 mm,5μm）,流动相为甲醇-0.2%磷酸（18∶82）;流速1.0 m L/min;检测波长250 nm;柱温30℃;进样量10μL。理论板数按葛根素峰计算应不得低于3 000。结果：葛根素对照品进样量在0.020 02~1.000 84μg范围内,进样量与峰面积呈良好的线性关系（r=0.999 9）,葛根素平均加样回收率为97.45%,RSD为1.62%（n=6）。结论：本方法操作简便,准确可靠,专属性强,重现性好。适宜用于测定葛根温胆颗粒中葛根素的含量。

Objective：To establish a HPLC method for the content determination of puerarin in gegen wendan granules.Methods：Samples were determined by HPLC on the C18column（250 mm × 4.6 mm,5 μm）.The column temperature was at 30 ℃; the inject volume was 10 μL.The mobile phase was consisted of methanol-0.2%phosphoric acid（18∶82）.The flow rate was 1.0 m L/min at UV detection wavelength of 250 nm.The number of theoretical plate calculated by puerarin peak should be not less than 3 000.Results：There was a good linear relationship of puerarin within the range of 0.020 02 ~ 1.000 84 μg,r = 0.999 9.The average recovery of puerarin was 97.45%,RSD = 1.62%（n = 6）.Conclusion：The method is convenient and accurate with a good specificity and reproducibility,it is suitable for the content determination of puerarin in gegen wendan granules.
