抑癌基因FN及PTEN在肝细胞肝癌中的表达及临床意义探讨 被引量:6

摘要 目的探讨纤黏连蛋白(fibronectin,FN)和张力蛋白同源基因蛋白(phosphatase and tensin homology deleted on chromosome ten,PTEN)在原发性肝细胞肝癌(hepatocellular carcinomas,HCC)组织中的表达及与其临床病理特征间的关系。方法采用Western blot及免疫组化方法检测83例HCC组织及其癌旁组织、47例肝硬变组织和11例正常肝组织中FN及PTEN的表达情况,并分析FN及PTEN的表达与HCC临床病理特征间的关系。结果 HCC癌组织中FN蛋白表达阳性率明显高于癌旁组织、正常肝组织及肝硬变组织(P<0.05),而PTEN蛋白的表达结果与之相反(P<0.05);癌旁组织中FN蛋白表达阳性率亦高于肝硬变组织及正常肝组织(P<0.05),PTEN蛋白的表达结果与之相反(P<0.05)。在合并癌栓、伴淋巴结转移、AFP阳性及肿瘤病灶多发的HCC癌组织中FN蛋白表达阳性率较高(P<0.05),而FN蛋白在HCC癌组织中的表达与患者性别、年龄、HBs Ag状态、肿瘤分化程度及大小均无关(P>0.05)。在高-中分化HCC、合并癌栓、AFP阳性、伴淋巴结转移和肿瘤直径≥2 cm的HCC癌组织中PTEN蛋白表达阳性率较低(P<0.05),而PTEN蛋白在HCC癌组织中的表达与患者性别、年龄、Hbs Ag状态及肿瘤数目无关(P>0.05)。结论 FN和PTEN在HCC癌组织中异常表达,其在HCC的发生、发展和侵袭转移中可能起一定的促进或抑制作用。两者异常表达可作为HCC恶性程度和侵袭转移的分子生物学标志物。 Objective To explore the relationship between the expressions of fibronectin (FN) and phosphatase and tensin homology deleted on ehromosome ten (PTEN) in hepatocellular carcinoma (HCC) tissues and the clinical pathological features. Methods The expressions of FN and PTEN were detected by using Western blot and immunohis- tochemistry respectively in 83 HCC tissues and para-carcinoma tissues, 47 hepatic cirrhosis tissues and 11 normal hepatic tissues. The correlations between the expressions of FN and PTEN and the dinicopathologic features of HCC patients were analyzed. Results The positive expression rate of FN protein in HCC tissues was significantly higher than those in para- carcinoma tissue, normal hepatic tissue, and liver cirrhosis tissues (P〈0.05); meanwhile the expression of PTEN was oppo- site (P〈0.05). The positive expression rate of FN protein in para-carcinoma tissues was also obviously higher than that liver cirrhosis tissues and normal hepatic tissues (P〈0.05), meanwhile the expression of PTEN was opposite (/9〈0.05). The positive expression rate of FN protein was higher in HCC tissues with cancer embolus, lymphatic metastasis, positive AFP, and multiple tumor (P〈0.05), but there were no statistically significant differences in FN protein expression, gender, age, HBsAg, degree of tumor differentiation, and size of tumor (P〉0.05). The positive expression rate of PTEN was lower in HCC tissues with high-medium differentiation, cancer embolus, positive AFP, lymphatic metastasis, and tumordiameter ≥2 cm (P〈0.05), there were no statistically significant differences in PTEN expression, gender, age, HBsAg, and the number of tumor (P〉0.05). Conclusions The abnormal expressions of FN and PTEN in HCC tissues which may play a role in promoting or inhibiting occurrence, development, invasion, and metastasis of HCC. The abnormal expres- sions of both can be used as molecular biological markers for the malignant degree, invasion, and metastasis of HCC.
