摘要
目的分析转座子Tn4001和操纵子ica ADBC与临床分离表皮葡萄球菌耐氨基糖苷类抗生素的关系。方法收集表皮葡萄球菌临床分离株77株,利用纸片扩散法检测细菌4种氨基糖苷类抗生素(庆大霉素、链霉素、奈替米星和阿米卡星)药敏特征;构建Tn4001和ica ADBC特异性引物,利用PCR扩增技术检测77株细菌Tn4001和ica ADBC。结果77株细菌对氨基糖苷类抗生素耐药率较高,分别为庆大霉素:62.3%;阿米卡星:22.1%;链霉素:61.0%;奈替米星:23.4%。Tn4001的检出率为58.4%,45株Tn4001阳性菌中,对庆大霉素、阿米卡星、链霉素、奈替米星耐药率分别为93.3%、33.3%、88.9%、37.8%。ica ADBC的检出率为20.8%;16株阳性菌中,对庆大霉素耐药率为93.8%;对阿米卡星耐药率为62.5%;对链霉素耐药率为93.8%;对奈替米星耐药率为62.5%。结论临床分离表皮葡萄球菌操纵子ica ADBC阳性菌株中通常伴随有转座子Tn4001检出,携带ica ADBC或Tn4001的菌株对氨基糖苷类抗生素耐药率较高。
Objective To research the relation between icaADBC ,Tn4001 and the Staphylococcus epidermidis antibiotics to aminoglycosides. Methods A total of 77 clinical isolates were tested for the antimicrobial susceptibility of 4 different kinds of aminoglycosides,including gentamicin,streptomycin,netimicin and amikacin,by the agar diffusion method; the Tn4001 and icaADBC specific primers were constructed,and Tn4001 and icaADBC of 77 bacterial were detected by PCR amplification technology. Results The 77 Staphylococcus epidermidis isolates had high resistance to aminoglycosides,as the following,gentamicin was 62. 3%,amikacin was 22. 1%,streptomycin was 61. 0%,and netilmicin was 23. 4%. The detection rate of Tn4001 was 58. 4%; and among the 45 positive Tn4001 strains,the resistant rates to gentamicin was 93. 3%,to amikacin was 33. 3%,to streptomycin was 88. 9%,and to netimicin was 37. 8%. The detection rate of icaADBC was 20. 8%; among the 16 positive strains,the resistant rate to gentamicin was 93. 8%,to amikacin was 62. 5%,to streptomycin was 93. 8%,and to netilmicin was 62. 5%. Conclusion The detection of clinical positive strains of Staphylococcus epidermidis operon icaADBC was usually accompanied by transposon Tn4001 detection. And the clinical Staphylococcus epidermidis isolates with Tn4001 or icaADBC show high resistance to aminoglycosides.
