自噬介导人舌鳞癌细胞对顺铂耐药的作用及其机制研究 被引量:3

The Study on the Role of Human Tongue Squamous Cell Cancer Cells Resistance to Cisplatin Mediated Autophagy and Its Mechanism

摘要 观测自噬在人舌鳞癌细胞对顺铂(DDP)耐药产生中的作用及分子机制,并进一步研究抑制自噬是否能够逆转肿瘤细胞对DDP的耐药,MTT法检测舌鳞癌Tca8113细胞的存活率,western-blot和RT-PCR法分别检测各实验组细胞外信号调节激酶(ERK)及雷帕霉素靶蛋白(mTOR)的表达情况。结果显示DDP联合氯喹组较DDP组细胞存活率低,差异有统计学意义(P<0.05),蛋白ERK的表达显著降低,mTOR的表达显著升高。因此,自噬抑制剂氯喹能够提高顺铂对舌鳞癌细胞的敏感性,增强了DDP的抗肿瘤作用。 This paper observed autophagy in human tongue squamous carcinoma cells to resistance cisplatin ,and discussed molecular mechanism and further studied whether the inhibition of autophagy can reversal of tumor cells resistant to cisplatin .MTT assay was used to detect the tongue squamous cell carcinoma cell line Tca8113 cell survival rate ,Western blot and RT‐PCR were detected each experimental group of ERK and mTOR expression . The results showed that combined with cisplatin in the chloroquine group and cisplatin group cell survival rate is of P〈0 .05 ,so the difference was statistically significant .The ERK protein expression decreased significantly ,and the expression of mTOR significantly increased .Therefore , chloroquine can improve the sensitivity of cisplatin on tongue cancer cells , and enhance the antitumor effect of cisplatin .
