膝关节骨性关节炎模型大鼠相关生物标志物表达及意义 被引量:3

KNEE OSTEOARTHRITIS RAT MODELS: THE EXPRESSION OF RELATED BIOMARKERS AND ITS SIGNIFICANCE

摘要 目的探讨膝关节骨性关节炎(OA)模型大鼠骨关节炎进展的不同时期相关生物标志物的表达及意义。方法将24只Wistar大鼠随机分为对照组(CG组)和3个实验组(TG1组、TG2组和TG3组),每组6只大鼠。实验各组行手术切除右膝内侧半月板及内侧副韧带,分别于术后2、4、6周切取标本进行组织学检查及蛋白聚糖、软骨寡聚基质蛋白(COMP)测定;CG组仅切开关节囊,于术后2周切取标本进行组织学检查及相关生物学标志物测定。结果随着实验时间延长,各实验组大鼠骨性关节炎Chambers评分增加(F=160.558,q=7.219~29.564,P〈0.05),实验组各亚组蛋白聚糖含量逐渐降低(F=39.561,q=4.599~14.736,P〈0.05),而COMP含量逐渐增加(F=25.521,q=6.488~10.528,P〈0.05),各组之间差异有显著意义。结论血清中COMP及蛋白聚糖浓度的测定有助于OA的早期诊断、疾病进展监测和预后判断。 Objective To explore the expression of biological markers in different periods of knee osteoarthritis model in rats. Methods Twenty-four Wistar rats were equally randomized to four groups as control group and three experimental groupsTG1 group,TG2and TG3 group.The rats in the three experimental groups received resection of both medial meniscus and medial collateral ligament of the right knee,the samples were taken from the three groups,respectively-2,4and 6weeks after surgery-for histologic examination and detection of proteoglycane and cartilage oligomerization matrix protein(COMP).The rats in the control group received only incision of joint capsule,histologic examination and detection of related biomarkers done two weeks after surgery. Results Along with the aggravation of Chambers score for rat osteoarthritis(F=160.558,q=7.219-29.564,P0.05),the proteoglycan levels of the three experimental groups gradually reduced(F=39.561,q= 4.599-14.736,P0.05),and the COMP levels gradually increased(F=25.521,q=6.488-10.528,P0.05),the dfferences between the three groups were significant. Conclusion Detection of serum levels of cartilage oligomerization matrix protein and proteoglycan is conducive to an early diagnosis,monitoring progression,and prognostic prediction of osteoarthritis.
